CKD model: nephrectomy 5/6

C57BL/6J mice or Sprague Dawley rats are used for 5/6 nephrectomy model (5/6 Nx). 5/6 Nx is performed in 6-wk-old mice/rats through two stages. In the first stage, the left kidney is decapsulated and the upper and lower poles are eliminated by constriction with silk thread via ventral incision. After one week, the second surgery is performed to completely remove the right kidney via dorsal incision.
Metabolic cages to estimate renal function

Urine analysis is carried out to estimate renal function in mice or rats that are placed in standard metabolic cages during 48 hours. The first 24 hours are used for animal adaptation to the metabolic cages and urine is discarded. The urine collected in the last 24 hours is used for biochemistry analysis of parameters related to renal function as creatinuria or albuminuria, and related to mineral metabolism as PTH, vitamin D, phosphate, calcium, among others.
AKI model: Bilateral renal Ischemia/Reperfusion

C57BL/6J mice are used for bilateral renal ischemia/reperfusion model (I/R). I/R is performed in 12-14-wk-old mice by renal artery clamping using a small non-traumatic vascular clamp for 45 minutes followed by reperfusion.
AKI model: Folic Acid-Induced Acute Kidney Injury

AKI is induced using the folic acid (FA) nephropathy model commonly used to study the pathophysiology associated with AKI. C57BL/6J adult mice (12-14 weeks old) are injected with a single intraperitoneal (i.p.) overdose of 250 mg/kg FA.
